In a rat restraint-stress model, researchers reported that stress reduced beneficial (obligate) colon bacteria and increased opportunistic microbes, and that Semax at 50 and 150 microg/kg prevented these stress-induced shifts in the microbiota. They suggest the effect may involve both central actions and peripheral intestinal melanocortin receptors. This is an intriguing gut-microbiome angle on Semax, but it is a small preclinical study with culture-based microbiota measures and a specific stress paradigm. Dose-dependence, with only mid-doses effective, also warrants caution. It adds breadth, not clinical weight. These are preclinical findings; human data are needed before clinical conclusions can be drawn.

Plain-language abstract

Stress can disturb the balance of bacteria living in the gut. This rat study tested whether Semax could protect that balance during a stressful restraint procedure. Rats received Semax at several doses shortly before being stressed. Stress alone reduced the helpful, normally dominant gut bacteria and allowed more potentially harmful microbes to grow. Semax at two middle doses (50 and 150 micrograms per kilogram) prevented these stress-driven changes, keeping the gut bacterial community closer to normal. The researchers suggest Semax may do this partly through the brain and partly by acting on receptors in the intestine itself. This is laboratory work in rats; it does not show an effect on the human gut, and only certain doses worked.
