[Normalizing effect of the pineal gland peptides on the daily melatonin rhythm in old monkeys and elderly people].

Among the more clinically relevant items in the epitalon literature, this report describes aged monkeys and elderly people, in whom night-time and average melatonin and circadian amplitude decline with age. The authors report that pineal preparations — epithalamin (a peptide complex) and epitalon — restored night-time melatonin release and normalised its daily rhythm, with elderly people who had low pineal function showing higher night melatonin. The abstract states no side effects were observed and frames a possible geriatric use. Weight this cautiously: it reads as an uncontrolled observational report, with no randomisation, blinding, sample size, or effect sizes given, from the single research group that dominates this field and whose methodology is difficult to assess in translation. The melatonin-normalisation hypothesis is plausible, but the human evidence here remains preliminary and hypothesis-generating.

As people and monkeys age, their bodies make less melatonin — the sleep-and-body-clock hormone produced by the pineal gland — especially at night, and its daily rhythm flattens. In this report, researchers gave two pineal-gland peptide preparations, epithalamin (a mix of peptides) and epitalon (a synthetic four-amino-acid peptide), to aged monkeys and elderly people. They reported that the preparations restored night-time melatonin release and helped return the hormone's daily cycle toward normal; elderly people whose pineal glands were underactive showed a rise in night-time melatonin. The authors stated they observed no side effects and suggested the peptides might have a role in care of older adults. Importantly, the abstract does not describe a comparison group, randomisation, or the number of participants, so these findings are preliminary and cannot establish cause and effect. This early human and animal work would need rigorous controlled trials to confirm.
