Overview of Epitalon-Highly Bioactive Pineal Tetrapeptide with Promising Properties.

A narrative review synthesizing roughly 25 years of Epitalon research — the tetrapeptide AEDG, derived from a bovine pineal-gland extract. It usefully catalogs the reported effects (geroprotective, neuroendocrine, antioxidant, neuroprotective, antimutagenic) and candidate mechanisms, from telomerase activation to influence on melatonin synthesis and immune signaling, and candidly flags how much remains uncertain — including that the compound's physico-chemical and structural characterization is still thin. As a review it aggregates but does not weigh study quality, and the underlying literature is dominated by in-vitro and animal work, much of it from a single research lineage; human clinical evidence is limited. So while the breadth of reported activity is striking, it should be read as a map of hypotheses, not a verdict on efficacy or safety. This review summarizes about 25 years of research on Epitalon (also spelled Epithalon or Epithalone), a peptide made of four amino acids (AEDG). It was originally created by copying the amino acids found in a natural extract of the pineal gland, a small gland in the brain. The authors gather findings from cell studies, animal studies, and computer-modeling studies. They report that Epitalon has been described as having anti-aging and hormone-related effects, thought to come from antioxidant, nerve-protecting, and anti-mutation actions. Reported mechanisms include boosting the enzyme telomerase, influencing production of the sleep hormone melatonin, and affecting immune-signaling molecules and various enzymes. The authors emphasize it is still unclear whether these are the only ways it works, and they point out that relatively little research has examined the peptide's basic chemical and physical structure. The review aims to pull together the most important findings and describe the current state of knowledge, rather than to prove that Epitalon works or is safe in people.
