A biodegradable suction patch for sustainable transbuccal peptide delivery.

This is a preclinical drug-delivery study, and bremelanotide and semaglutide are payloads used to test a device rather than the object of any efficacy question. The team built a biodegradable version of a bioinspired "suction patch" that sticks to the inner cheek to push peptides across the buccal lining — an attempt to avoid injections and gut degradation. In beagle dogs, semaglutide absorption was substantially better than the marketed tablet after a 10-minute application; bremelanotide reached about 26% of the bioavailability of a subcutaneous injection. Testing was ex vivo (porcine cheek tissue) and in animals, with no human data, so these are engineering and pharmacokinetic signals, not evidence of clinical benefit for either peptide's indication. The result says nothing about whether either drug works better delivered this way, only that meaningful amounts can cross the tissue. These are preclinical findings; human data are needed before clinical conclusions can be drawn about the delivery route.

Peptide drugs are hard to give as pills because stomach and gut enzymes break them down and their large size makes them hard to absorb, so many must be injected. This study tested a small patch that attaches to the inside of the cheek using gentle suction and helps peptides pass through the cheek lining into the bloodstream. Earlier versions of this patch used non-degradable materials; here the researchers rebuilt it from biodegradable plastics so it breaks down as waste after use, reducing environmental impact. They tested the materials for strength and confirmed they degrade in water and simulated waste, then checked performance using pig cheek tissue in the lab and living beagle dogs. In the dogs, the diabetes and weight drug semaglutide was absorbed much better than its commercial tablet after just 10 minutes on the cheek, and the desire drug bremelanotide reached about a quarter (26%) of the absorption seen with an under-the-skin injection. This was an engineering and absorption study in animals and tissue samples, not a test in people, and it did not measure whether either drug improved any health outcome.
