Liposomes as Carriers of GHK-Cu Tripeptide for Cosmetic Application.

An in-vitro cosmetic-formulation study focused on how to carry GHK-Cu into skin, not on clinical outcomes. Researchers built anionic and cationic lecithin liposomes (~100 nm) and characterised loading, bilayer fluidity and encapsulation efficiency (about 32% and 20% respectively for the selected carriers). In enzyme assays, GHK-Cu did not meaningfully affect tyrosinase but produced roughly 49% inhibition of elastase, which the authors link to slower elastin breakdown. This sits squarely in GHK-Cu's best-supported domain — topical skin science — but the endpoints are biochemical assays in a dish, not measurements on skin, animals or people. Encapsulation efficiency was modest, and no efficacy endpoint such as firmness or wrinkles was tested. These are preclinical, in-vitro findings; human data are needed before clinical conclusions can be drawn about any skin benefit.

Liposomes are microscopic fat bubbles that can carry active ingredients into skin. Researchers used them to package GHK-Cu, a copper-binding peptide of interest in anti-aging skincare. They made two versions — negatively charged and positively charged — using a film-hydration method with freeze-thaw and extrusion steps, then measured how well each held the peptide and how fluid its outer layer was. The best carriers trapped about 32% (positive) and 20% (negative) of the peptide and formed small, stable bubbles about 100 nanometers across. In test-tube enzyme experiments, the peptide had little effect on tyrosinase (an enzyme tied to pigmentation) but blocked about 49% of elastase activity — an enzyme that breaks down elastin, the protein that keeps skin springy. The authors conclude liposomes can successfully carry this peptide and that reducing elastase could help preserve skin structure.
