This rat study examines whether a synthetic glucocorticoid (methylprednisolone) blunts ipamorelin's action. It did not: the acute GH response to ipamorelin was preserved despite steroid treatment. More notably, ipamorelin given alongside methylprednisolone reduced the body-weight loss the steroid otherwise caused (rats on steroid alone lost about 13.6 g; with ipamorelin, losses fell to roughly 2 g), with a parallel rise in IGF-I. The authors read this as the GH-IGF-I axis remaining responsive to ipamorelin under glucocorticoid stress. It is a mechanistically clean result but a small rodent experiment with surrogate endpoints (body weight, IGF-I) rather than any functional or clinical outcome, and glucocorticoid catabolism in rats is only a loose analogue of human steroid-associated muscle loss. For Ipamorelin, which has no controlled human catabolic-illness trials, this is a preclinical signal. Human data are needed before drawing conclusions.

Plain-language abstract

Steroid medicines such as methylprednisolone can cause weight and muscle loss. This study in rats asked whether the peptide ipamorelin still works when a steroid is on board. In a first experiment, eight days of steroid slowed the rats' weight gain but did not reduce the immediate growth-hormone rise triggered by ipamorelin. In a second experiment, rats recovering from surgery received steroid alone or steroid plus ipamorelin (injected into a vein four times a day for 10 days). Rats on steroid alone lost about 13.6 grams of body weight, while rats also given ipamorelin lost only about 2 grams, a significantly smaller loss, and their IGF-I levels (a marker linked to growth-hormone activity) rose. The researchers concluded that the steroid does not block the body's growth-hormone response to ipamorelin, and that repeated dosing raised IGF-I and reduced weight loss in these animals. This is an animal study; it does not show the same effect in people.
