Do growth hormone-releasing peptides act as ghrelin secretagogues?

This is a rat mechanistic study probing where growth-hormone-releasing peptides act. Using whole-body autoradiography, researchers found that GHRP-6 and the ipamorelin-derived compound NN703 accumulate in the glandular stomach, the site where ghrelin, the endogenous ligand for the GHS-R1a receptor these peptides target, is made. Surgically removing part of the GI tract blunted the GH response to GHRP-6 by 60-70% while leaving the GHRH response intact, which the authors read as evidence that ghrelin release contributes to how GHRPs raise GH. It is a clever design, but the finding is a mechanistic inference from a single rodent study, and the resection model is a blunt tool. For Ipamorelin and GHRP-6 this adds texture to the receptor story rather than any clinical claim. These are preclinical findings; human data are needed before clinical conclusions can be drawn.

Researchers gave rats a radioactively labelled peptide and used whole-body imaging to see where it collected. A large amount gathered in the glandular part of the stomach, which is where the body makes ghrelin, a natural hormone that switches on the same receptor these peptides target. When part of the gut was surgically removed, the growth-hormone rise triggered by GHRP-6 dropped by about 60-70%, while the response to a different trigger (GHRH) was unchanged. The researchers concluded that some of the growth-hormone-releasing effect of these peptides may work through the body's own ghrelin, not only through direct action on the pituitary gland. This is early animal research and does not test any effect in people.
