This rat study investigated what happens when one adrenal gland is surgically removed — a procedure called unilateral adrenalectomy — and whether BPC 157, a synthetic 15-amino-acid peptide derived from a human stomach protein, could counter the resulting physiological damage.
In rats observed at 15 minutes, 5 hours, and 24 hours after surgery, researchers documented severe consequences including vascular injury, bleeding and clotting across multiple organs (including the brain, heart, lungs, liver, kidneys, and gastrointestinal tract), dangerous heart rhythm disturbances, elevated blood pressure in the portal and caval veins, and dangerously low aortic blood pressure. The researchers also observed abnormalities in the adrenal gland itself, including fat depletion and congestion.
BPC 157, given by mouth at two doses (10 micrograms per kilogram and 10 nanograms per kilogram), was associated with significant reduction in these effects. Results showed attenuation or elimination of arrhythmias, multi-organ hemorrhage, thrombosis, and vascular pressure abnormalities. The study reported that collateral blood flow pathways were reactivated, and markers of oxidative stress (specifically malondialdehyde levels) were reduced. Nitric oxide-related enzymes (NOS1-3) and a vascular growth factor (VEGF-A) were upregulated. Cortisol levels were higher than in untreated animals but remained very low overall.
