This controlled rodent study reports that semaglutide, alone and combined with PYY3-36, is associated with reduced hepatic inflammation and improved mitochondrial measures in a MASLD model. As preclinical work, the mechanistic findings should not be extrapolated to humans without clinical confirmation. The additive effect beyond food restriction is a noteworthy early signal for combination approaches.

In obese rats fed a fatty, high-fructose diet, semaglutide and the gut hormone PYY3-36 each reduced liver inflammation and fatty-liver severity, with additive benefits when combined. The combination also improved markers of mitochondrial function beyond what weight loss alone achieved.
