A PRISMA-registered systematic review and meta-analysis pooling seven RCTs on GLP-1 receptor agonists for antipsychotic-associated metabolic dysfunction. Weight and glycemic signals reached significance, but very high heterogeneity (I-squared=99%), small samples, and low-to-moderate GRADE certainty temper the conclusions. Readers can read the results as an early, cautiously encouraging signal that calls for larger, longer trials before firmer statements are warranted.
Plain-language abstract
This meta-analysis of seven randomized trials (446 participants) found that GLP-1 receptor agonists, including semaglutide, were associated with reductions in body weight, BMI, waist circumference, and fasting glucose in adults with schizophrenia taking antipsychotics. Effects on lipids, insulin measures, and blood pressure were inconsistent, and the overall certainty of evidence was low to moderate.
